Guitars are "near-line" level, but they're a high impedance output
with a series inductance. If the DI box (or any) input is less than
high impedance, it will definitely reduce the highs from a guitar.
This is why FET buffers and other high-impedance inputs are popular
for guitar, to keep full frequency response. Synths are "standard"
low-impedance (approximately) line-level output, so they should be
able to drive any input without a problem.

A DI box that uses a transformer for isolation does two things:

breaks the ground connection (provides isolation) between synth and
what it's plugged into, which can eliminate hum if it's caused by a
ground loop.

Adds a subtle (or less subtle, depending on how well you can hear it)
distortion to the sound. Many/most people consider this a "good"
distortion.
